Is your site hosted on a VPS? Or is it shared hosting? If server response time is the biggest issue, then a VPS can have the configuration modified to help improve server response time. This may be your greatest challenge. Beyond that, specific to the site itself, I can make sure compression is being used to speed up the transfer of files/information from the server to your visitors' browser. Also, I can make sure file caching is implemented, as well as asynchronous loading of included files. Lastly, I would setup a regularly scheduled job to optimize your database tables to avoid overhead/errors that can delay response times. All this would be measured via Google Insights.
